蜂螨实验(续)

I arranged mites for injections in the mornig. injected 100 mites, 50 GFP, 50 dsRNA for Sodium channel gene.   had to pick individual mites, stuck them on the ventral side with honey on a slide, and then a postdoc (another lab) injecated them under a scope. post injection, I took them down and washed each mite off honey with distilled water.  took 2 hours.

Two students looked for brood of the right ages in rain yesterday and got 4 of them. put them on my home yard around 3 pm.
Woke up around 4:30am even though my alarm was set at 6:30am. Got up at 6, too dark to do anything. 7 am still dark! waited till around 8 am. Used transparency sheets to make some brood that I thought be capped soon.  done around 8:40am.

went to campus at 10 am and injected 100 more mites, 50 GFP, 50 dsRNA for Sodium channel gene.  took only one hr today. back to home to eat lunch at 1 pm. then looked for brood for tomorow's rep. 2:30 pm, map the brood again using the same sheet to know which cells were capped within the last 6 hours. only in these cells can mite reproduce.  drove to lab. start mite transfer at 3:40 pm. done at 7 pm. had to open a cell that was capped in the designated time, put a mite in, then cover the hole, and seal with melted beeswax. some mites did not cooperate and would run around and to chase for a long time...

will put photos here and use chinese next week.
right now: 8:45pm. will try watch fast and furious 4. need to get up at 7am tomorrow morning to repeat the same. kicking myself that i did not do this earlier (like the 2nd week after I came back from China). now really cold. swithed from cool to heat on my central, and it started heating last night!  low around 48 last night.
